Drug–laboratory interaction between beta-lactam antibiotics and the galactomannan antigen test used to detect mould infections BJID

Several studies have demonstrated that piperacillin/tazobactam produces a false-positive result for the galactomannan antigen test. However, the most recent literature has demonstrated that this interaction is no longer a concern. There is little information regarding the drug–laboratory interaction with the generics of piperacillin/tazobactam or other broad-spectrum beta-lactams, such as ceftaroline, doripenem, imipenem/cilastatin, and meropenem. The purpose of this study was to determine if a drug–laboratory interaction exists with these antibiotics. Effect of abscisic acid on the mobilisation of galactomannan and embryo development of Sesbania virgata (Cav.) Pers. (Leguminosae - Faboideae) Rev. Bras. Bot.

Galactomannans (GM) are storage cell wall polysaccharides present in endospermic seeds of legumes. They are thought to be storage polymers, since it has been observed for a few species (among them Sesbania virgata) that they are completely broken down after germination and their products are transferred to the growing embryo. We examined the effect of 10-4 M abscisic acid (ABA) on the degradation of galactomannan in isolated endosperms and intact seeds of S. virgata. We found that after seed germination the initial embryo growth was retarded. Ultrastructural analysis showed that the embryo is completely surrounded by an endosperm which displays very thick galactomannan-containing cell walls. Galactomannans are storage cell wall polysaccharides present in seeds of some legumes. Their degradation is carried out by three hydrolases (alpha-galactosidase (EC 3.2.1.22), endo-beta-mannanase (EC 3.2.1.78) and ß-mannosidase (EC 3.2.1.25)). In the present study we purified and characterised an endo-beta-mannanase from seeds of Sesbania virgata and addressed its role in germination and seedling development. The polypeptide purified by Ion Exchange Chromatography and Affinity Chromatography on Sepharose-Concanavalin A, showed a pH optimum between 3.5 and 5 at 45ºC and high stability at pH 7.8. ABSTRACT Reserve mobilization in seeds occurs after visible germination, which is marked by the protrusion of the radicle or cotyledonary petiole, as in species of Arecaceae. Acrocomia aculeata (macaw palm), usually produces hard seeds whose endosperm has mannan-rich cell walls. We investigated the composition of storage compounds in macaw palm seed and the roles of two enzymes (endo-β-mannanase, α-galactosidase) during and after germination. The seeds were firstly submitted to pre-established protocol to overcome dormancy and promote germination. Galactomannan detection from piperacillin-tazobactam brands available in the Brazilian market BJID

Piperacillin-tazobactam is a broad spectrum antimicrobial agent that can cause false-positive results in the commercial Platelia Aspergillus EIA test. So far, no study has been performed in Latin America to evaluate the clinical implication of this finding. Here we studied the potential for galactomannan detection in piperacillin-tazobactam batches commercialized in the Brazilian market. Five batches from distinct laboratories were tested in duplicate in the Platelia Aspergillus EIA according to the manufacturer's instructions. Only one drug showed crossreaction at a cut-off of 0.5. Human serum was spiked with this particular drug aiming to mimic achievable piperacillin-tazobactam concentrations in the serum. Results were all negative for galactomannan...

Abstract Sunburn is responsible for significant losses in post-harvest and has motivated studies to diminish its effect on different types of cultivars. Thus, the present study proposes a rapid protocol to evaluate the photoprotective effect of suppressive formulations. The experiment consisted in placing fruits (mango) covered with different suppressor formulations about 7 centimeters from an incandescent source and measuring the surface temperature variation employing a thermometer and thermal camera.
